Drug mechanism
ACC inhibition.
Investigate a metabolic vulnerability through dual ACC1/ACC2 inhibition.
Published research informs the rationale. NMG formulation performance requires its own studies.
Our program
An ACC-inhibitor formulation for breast cancer, with improved therapeutic index as the development goal.
Where we are
We have built our first-generation ACC-inhibitor formulation and generated early preclinical data, including work in triple-negative breast cancer (TNBC) models. Formulation work and an exploratory animal pilot have informed follow-up decisions about dose, dosing frequency, and administration strategy.
Our current priority is a reproducible formulation for comparative testing. Planned studies will refine dose and schedule, evaluate TROP2 targeting, and assess antitumor activity, exposure, and tolerability together.

Science film / Optional music
A foundation already built: our first-generation ACC-inhibitor formulation and early preclinical data. The film explains the delivery question, ongoing formulation refinement, and planned TROP2 targeting studies, using triple-negative breast cancer to introduce the targeting hypothesis. Our lead program is breast cancer.
TROP2 is also expressed in normal tissues. Its presence alone does not establish selective delivery. Targeting, activity, exposure, and tolerability require experimental validation.
The film opens with the foundation already built: NanomediGene has produced a first-generation ACC-inhibitor formulation in a biodegradable PLGA carrier and generated early preclinical data. Formulation refinement continues. The film then introduces triple-negative breast cancer through three test-result badges: ER-negative, PR-negative, and HER2-negative. TROP2 is a cell-surface protein expressed in many TNBC tumors, with levels that vary between tumors. A TROP2-directed antibody-drug conjugate illustrates a treatment approach used in advanced TNBC.
The next diagram explains why drug distribution matters: free drug can reach tumors, bone marrow, and other tissues. Systemic ACC inhibition can affect platelet production. Adding a TROP2-binding ligand to NMG’s formulation is planned; whether it improves tumor-cell binding and uptake remains to be evaluated. Improved tumor delivery and reduced exposure elsewhere are goals, not demonstrated outcomes.
The next steps build on the existing formulation: improve reproducibility, evaluate TROP2 targeting, and assess antitumor activity and platelet effects in further preclinical studies. These are planned studies, not results. The conceptual animation does not establish the formulation’s safety, efficacy, or targeting performance.
Development priorities
Improve consistency of particle characteristics, loading, release, and stability.
Decision supported: A reproducible formulation suitable for comparative testing.
Evaluate ligand conjugation and TROP2-dependent binding and uptake.
Decision supported: Whether targeting adds value to the formulation.
Compare antitumor activity, drug exposure and distribution, and tolerability using defined active-drug doses and schedules.
Decision supported: A substantiated product profile and candidate-selection decision.
Develop manufacturing controls and the nonclinical package needed to support regulatory discussions.
Decision supported: Readiness to assess a potential IND submission.
Future work depends on study outcomes, resources, and regulatory requirements. This sequence does not represent a committed clinical start date.
Our approach
A metabolic mechanism, a formulation strategy, and a targeting question—each with its own evidence requirements.
Drug mechanism
Investigate a metabolic vulnerability through dual ACC1/ACC2 inhibition.
Published research informs the rationale. NMG formulation performance requires its own studies.
Formulation
Develop a reproducible formulation and test its effects on exposure, antitumor activity, and tolerability.
In development. Improved therapeutic index is the goal.
Planned targeting
Test whether adding a TROP2-directed ligand improves binding, uptake, and formulation performance.
Planned. Targeting remains to be validated.
Conceptual illustration of the development strategy; not experimental results. Prior studies of the free compound and other products do not establish the performance of NMG’s formulation.
Pilot learning / Next decisions
The exploratory pilot provided a starting point for follow-up studies of dose, frequency, and administration.
Prioritize a working dose range for systematic evaluation.
Use the pilot’s schedule observations to design controlled comparisons.
Use experience with administration routes to inform the next studies.
Planned formulation comparisons
Planned comparisons will include free-drug controls matched to the formulation’s active-drug amount per administration and dosing schedule. Administration route will be accounted for explicitly in study design and interpretation.
Administered dose and measured drug exposure are distinct. These studies are intended to establish the formulation’s contribution; the pilot did not establish an optimized regimen.
How we will evaluate therapeutic index
Human-relevant evaluations of platelet-producing cells (megakaryocytes) are under consideration. Platelet effects differ across species, so animal tolerability alone cannot establish human platelet safety. Published platelet research.
Planned evaluations; improved therapeutic index and platelet-sparing benefits remain to be demonstrated.
